pH sensor-reporter device in E. coli

This device consists of two plasmids. The first contains the ArsS/ArsR two-component pH sensor system. ArsS is a sensor kinase, and upon detecting an acidic environment, it phosphorylates ArsR, a transcription factor. The activated ArsR then binds to the pUreA promoter on the second plasmid, the reporter plasmid. ArsR binding activates expression of Comet GFP. The fluorescence of GFP corresponds means the cell is in an acidic environment, with the amount of fluorescence at a maximum for the optimal pH of the system (see Experience section).
